Disposable 24 Well Plate for Online pH Monitoring

HydroDish® HD24

The HydroDish® HD24 is a disposable polystyrene low-well multidish. Pre-calibrated pH sensors are integrated at the bottom of each round well and are read out contactless with the SDR SensorDish® Reader through the transparent bottom of the dish. They are delivered beta-irradiated.

Applications

Process Monitoring with the SDR in Suspension-Adapted CHO Cell Cultures

Oxygen and pH kinetics of suspension-adapted CHO cells at different start concentrations were monitored. The kinetics correlated well with the initial cell concentrations. pH values decreased only to a minimum of 6.3 due to daily media change. Samples with the lowest cell concentration did not show any pH change due to lack of cell growth. The oxygen uptake rate increased at each medium change. For the highest initial cell concentration it decreased at the end of the cultivation due to forming of aggregates. The results were confirmed by microscopic investigation.

pH Monitoring in 3D Culture of Disc-Chodrocytes

Hydrogel-based 3D cell culture systems offer particular advantages for tissue-engineering in
cell-based therapy for disc-degeneration. pH monitoring at different start cell densities was conducted in HydroDishes®. pH kinetics revealed characteristic dynamics according to the length of cultivation and the initial cell density. The pH values decreased without falling below pH 7.0 (Crabtree effect). At higher initial cell densities it took only about one day in order to reach the respective pH minimum. Due to the change of culture medium, a brief pH shift beyond physiologically optimal pH values took place. Removing the culture from the controlled incubator atmosphere (indicated by *) also resulted in a short increase in pH value, which could be recorded with the SDR.
